整合Arduino控制板之攜帶式CFPCR裝置

(Portable CFPCR chip device integrated with Arduino boards)

本組裝置具有小型對稱等溫區的微流體PCR晶片,只需一塊Arduino MEGA板即可進行溫度控制。另外,還組裝了一個微型注射幫浦,並編寫了一個Arduino程式,將樣品混合物注入晶片以實現PCR過程。

 

本裝置透過擴增200 bp的阿膠DNA片段,評估了各種操作參數下的PCR性能,以0.4μL/min的流速完成PCR過程所需的初始濃度為20 ng/μL。

 

我們的團隊是第一個將Arduino控制板導入CFPCR設備的溫度控制和樣品驅動模組的團隊。

This work describes the development of a portable nucleic acid amplification device that integrates thermal control and liquid pumping with Arduino boards. We have created a microfluidic PCR chip with small symmetric isothermal zones that require only one Arduino board for thermal control. We have also assembled a miniaturized liquid pump and programmed an Arduino file to push the sample mixture into the chip to implement the PCR process.

 

The sample flow's Nusselt number in our proposed operation is less than one, and heat transfer is conducted only. This ensures temperature uniformity in specific reaction regions. We have evaluated the PCR performance under various operational parameters by amplifying a Colla corii asini DNA segment of 200 bp. The initial concentration required to accomplish the PCR process is at least 20 ng/μL at the flow rate of 0.4 μL/min in the portable continuous flow PCR device.

 

Our group is the first to introduce Arduino boards into the heat control and sample pumping modules for a CFPCR device. Overall, this device can help identify traditional Chinese medicines, and its portability and affordability make it a valuable tool in various settings.
